DownloadLatestFileCharacterization of Bacillus subtilis surfaces by X-ray photoelectron spectroscopy. We have used X-ray photoelectron spectroscopy (XPS) to characterize the surfaces of Bacillus subtilis 168 and its pelE(c) mutant, which is a poor capsular polymer and therefore overproduces the polymeric amorphous polysaccharide known as exocellular polysaccharide (EPS). The XPS spectra were dominated by elements due to the outermost layers of the cell. Two distinct elements were detected: carbon with two components having binding energies at 284.6 and 289.4 eV, and nitrogen with two components having binding energies at 400.0 and 401.8 eV. There was no difference in the spectra between the wild type and the mutant, suggesting that the two components of the polysaccharide are a common constituent of the outermost layers of the cell. The carbon component was still detected in the pelE(c) mutant, suggesting that it is a constituent of the polymer rather than of the outermost layers of the cell. The nitrogen spectrum was still present in the mutant, suggesting that it is a constituent of some other outermost layer of the cell.The moment that Donald Trump was elected president, liberals weren’t just horrified, they were terrified.
