In this study, chromatin rich regions (CRR) on the surface of isolated mouse hepatocyte nuclei were studied by means of image analysis. The technique of isolated nucleus preparation yielded a mean yield of intact nuclei of 74.85% which was not significantly different from data obtained with other methods, but the chromatin density on the nuclei surface was significantly lower than that obtained with other methods. It is hypothesized that this decrease in chromatin density could be caused by the loss of tight chromatin structures during the isolation procedure and by elutriation. With isolated nuclei, image analysis revealed that the densities of CRR on the nuclei surface were higher in the low concentrations of 2 x 10(6) and 1 x 10(7) hepatocytes/ml than in higher concentrations. Two-minute incubation of 2 x 10(6) and 1 x 10(7) hepatocytes/ml resulted in significant decrease in density of CRR on the nuclei surface. The densities of CRR on the nuclei surface of intact and isolated nuclei were measured using a real-time mode of the whole field. This new technique may allow a more reliable measurement of CRR on the nuclei surface and may be helpful
